This study investigated the role of post-transcriptional regulation of gene expression in nerve inflammation associated with experimental autoimmune encephalomyelitis (EAE) in mice, by focusing on Hur, the RNA binding protein is involved in inflammatory and immune phenomena.

Intrathecal administration of anti-Hur antisense oligonucleotide (ASO) reduces the proinflammatory activated microglia inflammatory infiltrates and expression of proinflammatory cytokines IL-1β, TNF-α, and IL-17 and inhibits the activation of NF-kB road. The beneficial effects of anti-Hur ASO in EAE mice is also related to decreased permeability of the blood-brain barrier.
EAE mice showed decrease spinal CD206 immunostaining restored by anti-Hur ASO, showed that silencing Hur promote a shift to the anti-inflammatory and regenerative microglia phenotype. Mice that received anti-Hur related EAE ASO exhibited improved motor dysfunction, pain hypersensitivity, and body weight loss. Hur is targeting may represent an innovative and promising perspectives for the control of neurological disorders in patients with MS.

Acetylation / deacetylation and microtubule associated protein affects the stability and motility of sperm flagellar axonemal
PTM and MAPS is known to regulate microtubule dynamicity in somatic cells. Reported literature on α tubulin modulation acetyl transferase (αTAT1) and Histone Deacetylase 6 (HDAC6) in animal models and cell lines illustrates the differences in tubulin acetylation status associated with stability MT.
Our previous studies showed a decrease in sperm individual tubulin acetyl asthenozoospermic. Our study in mice sperm showed that the inhibition of HDAC6 activity despite increased tubulin acetylation, reduced sperm motility.
The study was therefore conducted to investigate the effect of tubulin acetylation / deacetylation in MT dynamicity in sperm flagella using rat and human sperm. Our data on mice revealed that sperm particular HDAC6 inhibitor Tubastatin A (T) inhibited sperm motility and neutralize the effects of debilitating Nocodazole depolymerizing and motility.
The effect on the polymerization was further confirmed in vitro using pure MT and recHDAC6. Also axoneme polymerized lacking in sperm from asthenozoosperm compared with normozoosperm.
Deacetylase Activity decreased in lysates sperm- and axonemes exposed T and N + T but not in sperm axonemes treated equally show that HDAC6 is associated with sperm axonemes or MT. Deacetylase Activity lacking in asthenozoosperm.
Interestingly, the expression of HDAC6 MDP3 physiologically known to bind to and inhibit its deacetylase activity, remained unchanged. However, the expression of acetyl-α tubulin, HDAC6 and stabilize microtubule protein lacking in asthenozoosperm SAXO1.
